Mazdutide dosage and protocol
Mazdutide is a dual GLP-1/glucagon receptor agonist being studied for metabolic research and weight management. It combines the mechanisms of both receptor systems.
- Vial size: 10 mg
- Reconstitution: 3 mL bacteriostatic water
- Concentration: 3.33 mg/mL
- Administration: subcutaneous
Research areas
- Metabolic Research
- Weight Management
- Glucose Regulation
Quick start
- Reconstitute: Add 3 mL bacteriostatic water to the 10 mg vial for about 3.33 mg/mL
- Typical dose: 3 mg once weekly for 4 weeks, then 6 mg once weekly
- Easy measuring: At 3.33 mg/mL, 1 unit is about 33.3 mcg on a U-100 syringe; 3 mg is about 90 units and 6 mg about 180 units (two draws)
- Storage: Lyophilized at -20°C; refrigerate after mixing and use within 28 days
Dosing protocol
| Phase | Dose | Frequency | Volume |
|---|---|---|---|
| Initiation | 3000 mcg (3 mg) | Once weekly | 0.60 mL |
| Maintenance | 6000 mcg (6 mg) | Once weekly | 1.20 mL |
How it works
Mazdutide (also known as LY3305677 or IBI362) is a once-weekly dual agonist of the GLP-1 receptor and the glucagon receptor, modeled on the oxyntomodulin hormone. The GLP-1 component increases glucose-dependent insulin secretion, slows gastric emptying and reduces appetite. The glucagon component adds effects on the liver and energy expenditure: in preclinical work glucagon receptor activation increased fat oxidation and energy use and reduced liver fat, while the GLP-1 activity offsets glucagon's tendency to raise blood glucose. Mazdutide was developed by Eli Lilly and Innovent Biologics. In phase 2 and phase 3 trials in Chinese adults with overweight or obesity, weekly mazdutide produced substantial weight loss compared with placebo, along with reductions in waist circumference, blood pressure, triglycerides and liver fat. Trials in type 2 diabetes showed HbA1c lowering. Based on these data it received regulatory approval in China in 2025 for chronic weight management; it is not approved in the United States or European Union. Long-term outcome data, including cardiovascular outcomes and head-to-head comparisons with other incretin drugs, are still limited.
Reconstitution steps
- Draw 2.0 mL bacteriostatic water with a sterile syringe
- Inject slowly down the vial wall; avoid foaming
- Gently swirl/roll until dissolved (do not shake)
- Label with reconstitution date and refrigerate at 2–8°C, protected from light; use within 28 days
Storage
Store lyophilized at -20°C. Refrigerate after reconstitution. Use within 28 days.
