FGL dosage and protocol
Neural Cell Adhesion Molecule peptide that enhances neuron growth and synaptic formation
- Vial size: 5 mg
- Reconstitution: 5 mL bacteriostatic water
- Concentration: 1mg/mL
- Administration: subcutaneous
Research areas
- Neurogenesis
- Memory Enhancement
- Learning
- Cognitive Function
- TBI Recovery
Quick start
- Reconstitute: Add 5 mL bacteriostatic water to the 5 mg vial for 1 mg/mL
- Typical dose: 1,000–2,000 mcg once daily for up to 8 weeks
- Easy measuring: At 1 mg/mL, 1 unit (0.01 mL) is 10 mcg on a U-100 syringe, so 1,000 mcg is 100 units (a full 1 mL syringe) and 2,000 mcg is 200 units
- Storage: Unreconstituted at −20°C; refrigerate at 2–8°C after mixing and use within 30 days
Dosing protocol
| Phase | Dose | Frequency | Volume |
|---|---|---|---|
| Standard | 1000-2000 | Once daily | 1-2 |
| Break | 0 | 2 days off weekly | N/A |
How it works
FGL (sometimes called FG Loop or FGLL) is a 15-amino-acid peptide derived from the second fibronectin type III module of the neural cell adhesion molecule (NCAM). NCAM normally signals partly through fibroblast growth factor receptor 1 (FGFR1); FGL mimics this interaction, binding and activating FGFR1 and downstream pathways including MAPK/ERK and PKC. These pathways regulate neurite outgrowth, synapse formation and neuronal survival. In cultured neurons, FGL stimulated neurite outgrowth, synaptogenesis and protection against apoptotic and amyloid-beta-induced toxicity. In rat studies, injected or intranasal FGL improved spatial and social memory, enhanced long-term potentiation in the hippocampus, reduced neuroinflammation, and in models of amyloid pathology lessened cognitive deficits and neuronal loss. Some work also explored cardioprotective effects. An early-phase human study explored intranasal FGL in Alzheimer's disease, but robust efficacy results have not been published, and FGL is not approved for any use. The evidence remains predominantly preclinical, and the subcutaneous doses in this record have not been tested in controlled human studies.
Reconstitution steps
- Draw 5.0 mL bacteriostatic water with a sterile syringe
- Inject slowly down the vial wall; avoid foaming
- Gently swirl/roll until dissolved (do not shake)
- Label and refrigerate at 2–8°C, protected from light; use within 30 days
Storage
Store unreconstituted at -20°C. Refrigerate after mixing at 2-8°C for up to 30 days
Important notes
- 5 days on, 2 days off protocol
- Younger subjects (30s) use lower 1mg dose
- Increases neuronal growth and synaptic formation
- Helps with traumatic brain injury recovery
- Antidepressant features
- Limited clinical safety data
Supplies
- Bacteriostatic water (5mL)
- Insulin syringes
- Alcohol swabs
