How it works

Dihexa (N-hexanoic-Tyr-Ile-(6) aminohexanoic amide) is a small, orally bioavailable peptide derivative of angiotensin IV developed at Washington State University. Rather than acting on classic angiotensin receptors, it binds hepatocyte growth factor (HGF) and promotes its dimerization, enhancing signaling through the HGF receptor c-Met. The HGF/c-Met system supports neuron survival, dendritic spine formation and synaptic connectivity. In rat studies, Dihexa improved performance in spatial learning tasks such as the Morris water maze in models of cognitive impairment, including scopolamine-induced deficits and aged animals. In cultured neurons it increased dendritic spine density and synapse formation. A frequently repeated claim that it is millions of times more potent than BDNF comes from a single in vitro assay of synaptogenic activity and does not translate into a demonstrated clinical comparison. There are no published controlled human trials of Dihexa. A related compound from the same program advanced to early clinical testing for Alzheimer's disease, but Dihexa itself remains a preclinical research chemical. Because c-Met signaling also drives growth and invasion in several cancers, there is a theoretical concern that sustained activation could promote tumor growth, and long-term safety is unknown.
