How it works

Argireline is the trade name for acetyl hexapeptide-3 (also listed as acetyl hexapeptide-8), a six-amino-acid peptide used in topical cosmetics. Its sequence mimics the N-terminal end of SNAP-25, one of the proteins that make up the SNARE complex required for nerve terminals to release acetylcholine. By competing with native SNAP-25 for a place in this complex, it is thought to destabilize SNARE assembly and modestly reduce neurotransmitter release at the neuromuscular junction. The intended downstream effect is reduced contraction of the small facial muscles responsible for expression lines, such as crow's feet and forehead lines. This is conceptually similar to botulinum toxin, which cleaves SNAP-25, but the effect is far weaker, fully reversible and depends on how much peptide actually penetrates the skin. Its penetration through intact skin is limited, which is a key uncertainty. Evidence comes from in vitro studies and small, often manufacturer-sponsored human cosmetic studies, which reported measurable reductions in wrinkle depth with twice-daily application over about four weeks. Independent, large controlled trials are lacking, and the effect is not comparable in magnitude to injectable neurotoxins. It is used topically, not by injection.
