How it works
PTD-DBM is a short peptide designed to interfere with the interaction between CXXC5 and Dishevelled (Dvl), two proteins that sit on the Wnt/β-catenin signalling pathway. In hair follicles, CXXC5 acts as a brake on Wnt signalling: it binds Dvl and dampens the pathway that drives follicle regrowth. PTD-DBM carries a protein-transduction domain (PTD) that lets it cross cell membranes, and a Dvl-binding motif (DBM) that competes with CXXC5 for Dvl. By occupying that site, it is intended to release the brake and restore β-catenin activity in follicle cells. The supporting evidence comes from a small number of laboratory and animal studies, mostly from one research group. In mice, topical or injected PTD-DBM was reported to promote hair regrowth after shaving and to speed wound healing, with effects described as additive to minoxidil in some experiments. This is a different mechanism from DHT-blocking drugs such as finasteride, which is why it is often described as acting on the follicle itself rather than on hormones. There are no published human clinical trials of PTD-DBM for hair loss that this reference is aware of, and products sold under the name vary in concentration and purity. Statements about human results should be treated as unproven, and the Wnt pathway is also involved in cell growth more broadly, so long-term safety has not been established.
