How it works
Selank is a synthetic heptapeptide (Thr-Lys-Pro-Arg-Pro-Gly-Pro) built from tuftsin, a fragment of immunoglobulin G, with a Pro-Gly-Pro tail added for stability. It was developed in Russia, where the standard form is approved as a nasal anxiolytic. Research suggests it acts through several routes: positive modulation of GABA-A receptor signaling, inhibition of enzymes that break down enkephalins (prolonging their effect), changes in serotonin and dopamine metabolism, increased BDNF expression in rodent brain, and modulation of immune signaling molecules such as IL-6. In Russian clinical studies, mostly small and published in Russian-language journals, Selank reduced anxiety symptoms in generalized anxiety and neurasthenia with effects compared to benzodiazepines but without notable sedation, withdrawal or memory impairment. Rodent studies show anxiety-reducing and memory-supporting effects. N-Acetyl Selank is a modified version in which the N-terminus is acetylated (and often the C-terminus amidated), intended to resist enzymatic breakdown and extend activity. This modification has not been evaluated in published controlled human trials; claims of improved potency or absorption are based on chemical reasoning and anecdote rather than direct evidence.
