How it works
FOXO4-DRI is a synthetic peptide built from D-amino acids in reverse order (a retro-inverso or D-Retro-Inverso design), which makes it resistant to protease breakdown. It mimics a region of the transcription factor FOXO4 and disrupts FOXO4's binding to the tumor suppressor p53 inside the nucleus of senescent cells. In senescent cells, this FOXO4-p53 interaction keeps p53 sequestered and prevents the cells from undergoing apoptosis. By blocking the interaction, FOXO4-DRI releases p53 to the mitochondria and triggers apoptosis selectively in senescent cells, while having little effect on normal proliferating cells in culture. Removing senescent cells reduces the pro-inflammatory secretions (the senescence-associated secretory phenotype) they produce. In a widely cited 2017 mouse study, intermittent FOXO4-DRI treatment restored fur density, physical fitness and kidney function in fast-aging and naturally aged mice and counteracted chemotherapy-induced toxicity. Later rodent work reported benefits in models such as age-related testosterone decline. No human clinical trials have been published, and the doses and schedule used in mice (high doses given every other day for short periods) differ substantially from continuous daily low-dose protocols. Human safety and efficacy are unknown.
Reconstitution steps
- Draw 3.0 mL bacteriostatic water with a sterile syringe
- Inject slowly down the vial wall; avoid foaming
- Gently swirl/roll until dissolved (do not shake)
- Label and refrigerate at 2–8°C, protected from light
Storage
Store lyophilized at −20°C. After reconstitution, refrigerate at 2–8°C and use within 1 month with bacteriostatic water. Avoid freeze–thaw cycles.
