Gonadorelin dosage and protocol
Gonadorelin is a synthetic decapeptide identical to endogenous gonadotropin-releasing hormone (GnRH) secreted by the hypothalamus. It stimulates the pituitary to release LH and FSH, supporting testosterone production in men and ovulation in women when administered in short, pulsatile doses. Commonly used to maintain testicular function and fertility during or after testosterone therapy.

- Administration: subcutaneous
Research areas
- Testosterone Support
- Fertility

Quick start
- Reconstitute: Add 2.0mL bacteriostatic water → 1mg/mL concentration
- Typical dose: 50–150mcg per injection, 2–3x weekly
- Easy measuring: At 1mg/mL, 1 unit = 0.01mL = 10mcg on a U-100 syringe
- Storage: Lyophilized at −20°C; reconstituted refrigerated at 2–8°C; use within 4 weeks
Dosing protocol
| Phase | Dose | Frequency | Volume |
|---|---|---|---|
| Weeks 1–2 (Initiation) | 50mcg | 2–3x weekly (non-consecutive days) | 5 units (0.05mL) |
| Weeks 3–4 (Titration) | 100mcg | 2–3x weekly (non-consecutive days) | 10 units (0.10mL) |
| Weeks 5–8 (Maintenance) | 100–150mcg | 2–3x weekly (non-consecutive days) | 10–15 units (0.10–0.15mL) |
How it works
Gonadorelin is a synthetic decapeptide identical to endogenous GnRH. When administered in short, pulsatile fashion, it binds to GnRH receptors on pituitary gonadotrophs, triggering the release of LH and FSH. LH stimulates Leydig cells to produce testosterone in men and triggers ovulation in women, while FSH supports spermatogenesis and ovarian follicle development. Unlike continuous GnRH agonist administration (which desensitizes receptors), brief periodic dosing preserves the natural pulsatile signaling pattern.
Reconstitution steps
- Draw 2.0mL bacteriostatic water with a sterile syringe
- Inject slowly down the vial wall; avoid foaming
- Gently swirl/roll until dissolved (do not shake)
- Label concentration (1mg/mL) and date; refrigerate at 2–8°C, protected from light
Storage
Lyophilized: store at −20°C (−4°F). After reconstitution, refrigerate at 2–8°C and use within 4 weeks. Avoid freeze–thaw cycles.
Important notes
- Research use only — not approved for general human use outside specific indications
- Limit continuous daily dosing to ≤14 consecutive days to prevent pituitary desensitization
- Administer on non-consecutive days to mimic natural pulsatile GnRH release
- Often used alongside testosterone therapy to maintain testicular size and fertility
- Monitor LH, FSH, and testosterone levels periodically to assess response
- Not suitable for those with hormone-sensitive conditions without medical supervision
Supplies
- Gonadorelin vial (2mg)
- Bacteriostatic water (10mL bottle)
- Insulin syringes (U-100, 30- or 50-unit preferred)
