Kisspeptin-10 dosage and protocol

Puberty peptide that stimulates sex hormone production and enhances fertility in both men and women

  • Vial size: 10 mg
  • Reconstitution: 3 mL bacteriostatic water
  • Concentration: 3.33 mg/mL
  • Administration: subcutaneous

Research areas

  • Testosterone Support
  • Fertility
  • Sexual Health
  • Hormone Optimization

Quick start

  • Reconstitute: Add 3 mL bacteriostatic water to the 5 mg vial → about 1.67 mg/mL
  • Typical dose: 100-200 mcg, 3-5 times weekly for 4 weeks, then 4 weeks off
  • Easy measuring: At 1.67 mg/mL, 1 unit = 0.01 mL ≈ 16.7 mcg on a U-100 syringe, so 100 mcg ≈ 6 units and 200 mcg ≈ 12 units
  • Storage: Unreconstituted at -20°C; refrigerate after mixing

Dosing protocol

Weeks 1–2100 mcgOnce daily (SC)0.03 mL (3 units)
Weeks 3–8200 mcgOnce daily (SC)0.06 mL (6 units)

How it works

Kisspeptin-10 is the shortest active form of kisspeptin, a family of peptides encoded by the KISS1 gene. It activates the kisspeptin receptor (KISS1R, formerly GPR54) on gonadotropin-releasing hormone (GnRH) neurons in the hypothalamus. This triggers GnRH release, which in turn prompts the pituitary to release luteinizing hormone (LH) and, to a lesser extent, follicle-stimulating hormone (FSH). LH and FSH then drive testosterone production and sperm development in men, and ovarian hormone production and ovulation in women. Loss-of-function mutations in the receptor cause failure of puberty, which is how the system's importance was discovered. In small human clinical studies, injected kisspeptin-10 rapidly raised LH and testosterone in healthy men, and it has been studied in people with low reproductive hormone levels. The longer form, kisspeptin-54, has been used experimentally to trigger egg maturation in IVF trials with a reportedly lower risk of ovarian hyperstimulation than standard triggers. Kisspeptin-10 has a very short half-life (minutes), and continuous exposure can desensitize the system and suppress, rather than stimulate, hormone release. It is not an approved medicine, and evidence is limited to short-term research studies.

Reconstitution steps

  • Allow to room temperature
  • Add 3mL bacteriostatic water
  • Swirl gently
  • Refrigerate

Storage

Store unreconstituted at -20°C. Refrigerate after mixing

Important notes

  • Stimulates GnRH release in hypothalamus
  • Releases LH and FSH in pituitary
  • Men: LH/FSH signal testes to produce testosterone and sperm
  • Women: LH/FSH signal ovaries to produce estrogen and trigger ovulation
  • 1mcg per kg body weight (100-200mcg typical)
  • Best in fasted state in morning for natural testosterone rhythm
  • Peak FSH & LH occurs 45-60 minutes after injection
  • 3-5x week, 1 month on, 1 month off
  • For fertility: take 45-60 mins before conception attempts
  • Temporarily increases oxytocin
  • Low cost with high benefit profile

Supplies

  • Bacteriostatic water (5mL)
  • Insulin syringes (0.5mL)
  • Alcohol swabs
