How it works
Leuphasyl (pentapeptide-18) is a cosmetic peptide designed to mimic the action of enkephalins, the body's own opioid-like signaling peptides. It is thought to bind enkephalin receptors on the surface of nerve cells near the neuromuscular junction, which reduces neuronal excitability and lowers the release of acetylcholine. Less acetylcholine reaching facial muscles means weaker repetitive contraction, the process behind expression lines such as crow's feet and forehead creases. This mechanism is upstream of, and complementary to, Argireline (acetyl hexapeptide-8), which interferes with the SNARE complex that releases acetylcholine. For that reason the two are frequently paired in formulations, and manufacturer-run studies have reported additive reductions in wrinkle depth when combined. The evidence base consists mainly of in vitro neuron studies and small, mostly manufacturer-sponsored cosmetic trials measuring wrinkle depth over several weeks. Effects reported are modest and far smaller than those of injected botulinum toxin, and independent peer-reviewed replication is limited. Leuphasyl is intended for topical use; it is not a systemic drug and has no established role as an injectable.
