SNAP-8 dosage and protocol
SNAP-8 (Acetyl Octapeptide-3) is a synthetic octapeptide cosmeceutical and extended analog of Argireline, designed to mimic the SNAP-25 protein involved in neuromuscular signaling. By competitively inhibiting SNAP-25 in the SNARE complex, it attenuates acetylcholine release at neuromuscular junctions, relaxing facial muscle contractions without causing paralysis. Clinical studies showed ~35% wrinkle reduction and improved skin elasticity with a favorable safety profile.

Quick start
- Reconstitute: Add 3.0 mL bacteriostatic water to the 10 mg vial for about 3.33 mg/mL
- Typical dose: 330-1000 mcg once daily, titrated over 12 weeks
- Easy measuring: At 3.33 mg/mL, 1 unit (0.01 mL) on a U-100 syringe is about 33 mcg; 330 mcg is about 10 units and 1000 mcg is 30 units
- Storage: Lyophilized at -20°C, dry and dark; refrigerate at 2-8°C after reconstitution and use within 30 days
Dosing protocol
| Phase | Dose | Frequency | Volume |
|---|---|---|---|
| Weeks 1–4 | 330mcg | Once daily | 0.10mL |
| Weeks 5–8 | 500mcg | Once daily | 0.15mL |
| Weeks 9–12 | 1000mcg | Once daily | 0.30mL |

How it works
SNAP-8 (acetyl glutamyl heptapeptide-3, often sold as acetyl octapeptide-3) is a cosmetic peptide that mimics the N-terminal end of SNAP-25, one of the proteins that forms the SNARE complex. The SNARE complex is needed for vesicles carrying acetylcholine to fuse with the nerve terminal membrane. By competing with SNAP-25, SNAP-8 is proposed to destabilise SNARE assembly and reduce acetylcholine release, softening muscle contraction rather than blocking it the way botulinum toxin does. The intended downstream effect is reduced depth of expression lines such as forehead and crow's feet wrinkles. In vitro work showed reduced SNARE complex formation and neurotransmitter release in cell models. Evidence for wrinkle reduction comes mainly from small, manufacturer-sponsored studies of topical creams, and independent peer-reviewed data are limited. SNAP-8 was developed and studied as a topical cosmetic ingredient; there are no published safety or efficacy studies of subcutaneous injection, which is the administration route listed in this record. Skin penetration of topical peptides of this size is also debated.
Reconstitution steps
- Draw 3.0mL bacteriostatic water with a sterile syringe
- Inject slowly down the vial wall; avoid foaming
- Gently swirl/roll until dissolved (do not shake)
- Label and refrigerate at 2–8°C, protected from light
Storage
Store lyophilized at −20°C (−4°F) in dry, dark conditions. After reconstitution, refrigerate at 2–8°C; use within 30 days for maximum potency. Avoid freeze–thaw cycles.
Important notes
- Research use only
- Extended analog of Argireline with ~30% greater activity in reducing neurotransmitter release
- Unlike Botox, provides gentle non-paralyzing relaxation that preserves natural facial expressions
- Results typically noticeable after 2–4 weeks of consistent daily use
- Rotate injection sites (abdomen, thighs, upper arms) to reduce local irritation
- Cycle length 8–12 weeks; optional extension to 16 weeks
Supplies
- Bacteriostatic water
- Insulin syringes (U-100)
- Alcohol swabs
